Methotrexate is a drug that inhibits folic acid synthesis. It is used as an anticancer drug and an immunosupressant.

Mechanism
- competitively and reversibly inhibits dihydrotetrafolate reductase (DHFR), an enzyme in the folate synthesis pathway
- Folate is required for thymidine synthesis
