- drug of choice only for Typhoid fever due to serious risk of aplastic anemia
- wide range of gram-negative and -positive bacteria
- active against Rickettsia, Chlamydia, Mycoplasma

- inhibits protein synthesis by binding the 50S ribosomal subunit
